Desarrollo de métodos de referencia para la cuantificación de citosinas metiladas en fragmentos de ADN

Abstract:
La metilación del ADN es una de las modificaciones epigenéticas más estudiadas en seres humanos dada su vital importancia en diferentes procesos de la regulación génica. En los últimos años se ha comprobado que las alteraciones en los patrones de metilación están fuertemente asociadas a numerosas enfermedades entre las que se incluye el cáncer, por lo que la metilación del ADN se está monitorizando como biomarcador de determinados tipos de tumores. Con el auge de la espectrometría de masas y las ventajas que esta técnica proporciona, se han comenzado a desarrollar metodologías analíticas basadas en cromatografía líquida acoplada a espectrometría de masas para la cuantificación de citosinas metiladas. En este trabajo se propone la puesta a punto de un método de referencia sencillo y preciso para la cuantificación de citosinas metiladas en fragmentos de ADN mediante HPLC-MS/MS y dilución isotópica. Durante el trabajo, se llevó a cabo la extracción de ADN en algas enriquecidas isotópicamente en 15N y su posterior hidrólisis para la conversión a nucleósidos, mucho más fáciles de analizar por espectrometría de masas que las moléculas completas de ADN. Para ello se optimizó la separación cromatográfica y su posterior detección en el MS/MS, además de realizar la caracterización mediante dilución isotópica inversa de los compuestos obtenidos marcados isotópicamente. Finalmente, se cuantificó mediante dilución isotópica el porcentaje de metilación de ADN procedente de dos tejidos de órganos diferentes de un patrón comercial de ratón, siendo ésta insatisfactoria e impidiendo así la validación del método desarrollado, lo que se puede deber a distintos motivos que se discutirán a lo largo del trabajo.
